Figure 7: P gingivalis upregulated TIMP-1 and TIMP-2 mRNA expression. Following osteoblast culture in the presence or absence of live and heat-inactivated P gingivalis (107 bacteria), total RNA was extracted from the osteoblasts and RT-PCRs were performed using specifi c primers for TIMP-1, TIMP-2, and GAPDH. Changes in mRNA levels were measured using the public domain NIH Image software package. P gingivalisinfected cultures and noninfected cultures (CTRL) were compared. CTRL represents 100% gene activation and was used to determine the relative expression levels of TIMPs mRNA. Photos are of representative gels of three separate experiments.
